Summary |
We used the Structure-seq2 protocol and applied it to 14-day-old rice (Oryza sativa) shoot tissue for genome-wide RNA structure probing to investigate the effect of heat stress on the RNA structurome.

Overall design |
We applied our newly optimized pipeline of Structure-seq2 on two weeks old rice shoot. 1 g of shoot tissue were collected right before each treatment. -/+DMS treatments (3 biological replicates for each) was applied for room temperature treatment as the control. For the heat shock treatment, −DMS and +DMS samples were similarly prepared but under 42°C. Total 12 individual libraries was labeled with unique Illumina TruSeq barcode and were sequenced by HiSeq2500.
