Summary |
We analyzed genome-wide chromatin binding of the transcription factor c-Myb using ChIP-Seq. K-562 cell lines stably expressing N-termianl 3xTY tagged Myb (pEFneo-3xTY-Myb) and control cell lines expressing the tag (pEFneo-3xTY) were used.

Overall design |
K-562 cells stably expressing pEFneo-3xTY-Myb and pooled control cell lines expressing pEFneo-TY were crosslinked followed by nuclei isolation and sonication. Sonicated chromatin was immunoprecipitated with a mouse monoclonal anti-TY1 antibody followed by elution of immnoprecipitated chromatin into two fractions. Pooled fractions were subjected to DNA purification using 'ChIP DNA clean concentrator KIT' (Zymo research #D5205). Libraries were prepared using ThruPLEXⓇ low-input sample-prep method. 75 bp single-end reads were generated from three biological replicates using the Illumina NextSeq 500 sequencer.
