Summary |

Overall design |
Breast organoids from patients without (normal) or with BRCA1 mutations were cultured in vitro. Organoids from each patient were divided into 2 groups. One group was treated with estradiol and progesterone and the second group was treated with estradiol, progesterone and TPA. Hormone treatment over the course of 28 days was as follows: 0.1nM estradiol on days 1-7, 1nM estradiol on days 7-14, 1nM estradiol + 10nM progesterone (with or without 1uM TPA) on days 14-21 and 0.1nM estradiol + 50nM progesterone (with or without 1uM TPA) on days 21-28. Media was changed every 2 days.
