Title BRCA1 mutation influences progesterone response in human benign mammary organoids.

Summary Mammary organoids treated with a 28 day menstrual cycle profile of estradiol and progesterone with or without TPA: Comparison of normal versus BRCA1 mut cells. Purpose: The goals of this study are to determine 1) the effects of a 28 day menstrual cycle profile of estradiol and progesterone, 2) the effects of TPA, and 3) the responses in normal versus BRCA1 carriers, in benign breast organoids using RNA-seq
 
Overall design Breast organoids from patients without (normal) or with BRCA1 mutations were cultured in vitro. Organoids from each patient were divided into 2 groups. One group was treated with estradiol and progesterone and the second group was treated with estradiol, progesterone and TPA. Hormone treatment over the course of 28 days was as follows: 0.1nM estradiol on days 1-7, 1nM estradiol on days 7-14, 1nM estradiol + 10nM progesterone (with or without 1uM TPA) on days 14-21 and 0.1nM estradiol + 50nM progesterone (with or without 1uM TPA) on days 21-28. Media was changed every 2 days.
