Summary microRNAs are crucial post-transcriptional regulators which contribute to the effect of sulforaphane in cancer and are also involved in the modulation of of many aspects of DC biology, including development, maturation, surface molecule expression and released cytokines
We used microarrays to detail the regulation of miRNAs in human DCs by sulforaphane, underlying the regulation of different crucial DCs regulatory molecules and signaling pathways
 
Overall design human monocyte-derived DCs (MoDCs) were produced, treated as described and used for miRNA extraction and hybridization on Affymetrix GeneChip miRNA 4.0 Arrays. We sought to increase the quality of produced MoDCs by isolation of initial Monocytes by CD14 MACS microbeads. The high quality MoDCs were proved by flow cytometry, pretreated with 30 mM sulforaphane (SF) for 30 min, treated with maturation cytokine cocktail or left untreated and collected for miRNA isolation after 48 hours incubation
