Overall design |
Protein A-Tn5 was enriched through specific antibodies to genomic regions and Tn5 generated indexed chromatin. Following tagmentation, mRNA inside cells were captured by oligo dT barcode primers and reversed-transcribed. Combinatorial indexing strategy was implemented on both ChIPed DNA parts and RNA parts.
