Overall design |
Determination of the sequence of full-length transcripts from mouse embryonic stem cells (ESC) cultured in naïve 2i conditions. PolyA-enriched mRNAs were selected by hybridization to a custom oligo probe set and post-capture, mRNAs were amplified using the Clontech SMARTer PCR cDNA Synthesis Kit with 9 cycles and used in the SMRTbell library prep. The library was sequenced on the SMRTseq 2000 platform. PacBio reads were processed through the SMRTLINK v8.0.0 IsoSeq3 pipeline and the PacBio Isoseq transcripts were mapped using minimap2.
