Title Global transcriptome analysis of long RNA molecules expressed in human keratinocyte cell line HaCaT pre-treated with TGF-β1 in response to silymarin, a natural plant extract involved in skin tissue regeneration

Summary The goal of this study is to find out the exact mechanisms of action of silymarin (SM), a complex mixture of flavonolignans structures isolated from milk thistle (Silybum marianum), on the biological behavior (phenotype) of human HaCaT keratinocytes.
 
Overall design To place the HaCaT cells in a context of a tissue re-epithelization process, we treated these cells for 24 hours with the Transforming Growth Factor β1 (TGF-β1, 10 ng/ml) in presence or absence of silymarin (SM, 31.25 µg/ml). Total RNA (longer than 200 nucleotides) were isolated, polyA RNA were enriched and sequenced from 3 biological replicate samples of TGF-β1-treated HaCaT cells (TGF) and 3 biological replicate samples of TGF-β1 plus SM-treated HaCaT cells (TGF.SM31).
