Title |
Macrophage heterogenity in liver and adipose during obesity |
Organism |
Mus musculus |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
Liver and adipose tissue undergo dynamic inflammatory changes during the progression of obesity. This study was designed to evaluate the changes in gene expression profile and function of subpecific macophage subsets.

Overall design |
Cx3Cr1-GFP mice were fed a STD or high fat, high sucrose diet for 16 weeks after which macrophages were isolated from the adipose tissue and liver. Flow sorting was utilized to divide liver macrophages into F4/80hi cells expressing TIM4 [i.e., STD KC, HFD KC] or those expressing Cx3cr1 (i.e., GFP positive) [Cx3cr1pos macs-HFD]. In the adipose tissue, high fat diet ATMs were flow sorted based on their expression of F4/80 and CD64 [i.e., ATMs-HFD]. A total of 3 samples from STD KC, 5 samples from HFD KCs, 4 samples of Cx3cr1 Macs, and 5 ATM samples on HFD were collected.
