Summary |
Human astrovirus infection is known to disrupt intestinal barrier function by increasing barrier permeabilty. However, the exact cellular mechanism(s) involved is unknown. We used microarrays to detail the global gene expression changes occuring during astrovirus infection and identify necessary cellular pathways for astrovirus pathogenesis.

Overall design |
Total RNA was extracted from human astrovirus HAstV-1 infected (24 hours post-infection) and non-infected Caco-2 cells and gene expression profiles were analyzed using Affymetrix Human Gene 2.0 sense transcript (ST) GeneChip microarrays. In order to determine genes modulated by infection and those required for infection we compared infected to non-infected Caco-2 cells as well as Cac0-2 cells that are permissive to infection (over-confluent) and non-permissive (sub-confluent) to infection. To perform this analysis, we harvested RNA from HAstV-1 infected (MOI of 10) and non-infected Caco-2 cells at 24 hours post-infection. Additionally, RNA was extracted from Caco-2 cells grown to differentiation which are permissive to HAstV-1 infection and cells grown only to sub-confluency which are non-permissive to HAstV-1 infection.
