Summary |
Liquid cultures of the unicellular green alga, Chlamydomonas reinhardtii were grown in media with 6 uM Mn (control) or 1000 uM Mn (experimental), and analyzed by RNA-Seq to identify genes that are differentially expressed in response to excess Mn.

Overall design |
Six flasks containing Tris-Acetate-Phosphorus (TAP) medium were innoculated with C. reinhardtii from a starter culture. Three of the flasks contained 6 uM Mn and three flasks contained 1000 uM Mn. These were grown on a shaker platform under continuous light until they reached a cell density of 3x10^6 cells/mL (mid-log). Total RNA was purified from each culture and subjected to RNA-Seq analysis on the Illumina HiSeq2000 platform.
