Title Transcriptomics study of excess Mn (1000 uM) in Chlamydomonas reinhardtii
Organism Chlamydomonas reinhardtii
Experiment type Expression profiling by high throughput sequencing
Summary Liquid cultures of the unicellular green alga, Chlamydomonas reinhardtii were grown in media with 6 uM Mn (control) or 1000 uM Mn (experimental), and analyzed by RNA-Seq to identify genes that are differentially expressed in response to excess Mn.

Overall design Six flasks containing Tris-Acetate-Phosphorus (TAP) medium were innoculated with C. reinhardtii from a starter culture. Three of the flasks contained 6 uM Mn and three flasks contained 1000 uM Mn. These were grown on a shaker platform under continuous light until they reached a cell density of 3x10^6 cells/mL (mid-log). Total RNA was purified from each culture and subjected to RNA-Seq analysis on the Illumina HiSeq2000 platform.
