Summary |
Recent studies have suggested that formation of supramolecular activation cluster (SMAC), a large ‘immunological synapse’ composed of various signaling complexes, enhances CD4+ T cell activation. Critically, although the composition of SMAC is known, the proteins involved in SMAC formation have not been fully investigated. We identified that intraflagellar transport 20 (IFT20) play central roles in the effector functino of CD4+ T cells.

Overall design |
We analyzed in vitro activated CD4 T cell by single cell RNA sequencing (scRNAseq) with 10X genomics. CD4+ T cells for T cell activation were extracted from spleens of Ift20f/f or Ift20f/f CD4cre mice. Following single cell isolation, the MagnisortTM Mouse CD4 naïve T cell Enrichment Kit (Invitrogen) was used to obtain purified naïve T cells. Naïve CD4 + T cells were activated with 1-μg/mL anti-CD3 (Invitrogen) and 1-μg/mL anti-CD28 antibodies (BioLegend) for 24 hours.
