Title Effect of SigA depletion and SigB knockout on the global transcription of Mtb
Organism Mycobacterium tuberculosis
Experiment type Expression profiling by high throughput sequencing
Summary Analyzing differences in transcriptome profile of RvΔsigA (SigA conditional knockdown strain) in presence of anhydrotetracycline and transcriptome profile of RvΔsigB (SigB knockout strain) compared to control.

Overall design RvΔsigA was inoculated at 0.05 OD, in presence or absence of 1mM/ml anhydrotetracycline. Post 3 days total RNA was islolated and RNA seq was performed. RvΔsigB was inoculated at 0.05 OD and grown to 0.8 OD, after which RNA was isolated and RNA sequencing was carried out.
