Title 'Effect of markerless deletion of PA14_16550 from P. aeruginosa PA14 biofilm colonies on M6301-glycerol agar for 3 d at 25C

Summary To determine the transcriptomic effect when PA14_16550, which encodes a TetR-type DNA-binding protein, is absent from cells. Deletion of this gene results in lower exopolysaccharide levels (a biofilm defect). Here, we compared PA14 deleted for amrZ (which has a moderately hyper-biofilm phenotype) and cells deleted for both amrZ and 16550. We also analyzed wild-type PA14 cells.
 
Overall design Total RNA was extracted from mechanically dispersed colonies grown on M6301-1% agar for 3 d at 25C. Transcriptomes were compared among PA14, PA14 delta-amrZ, and PA14 delta-amrZ delta-16550.
