Title Single-cell RNA-sequencing of stromal vascular cells (SVCs) from the epididymal fat of obese mice.

Summary The obesity epidemic continues to worsen worldwide, driving metabolic and chronic inflammatory diseases. Thiazolidinediones (TZDs), such as rosiglitazone (Rosi), are PPARy ligands that promote anti-inflammatory changes in adipose tissue macrophages (ATMs) and cause insulin sensitization. We performed single-cell RNA-seuqnecing (scRNA-seq) to analyze the diversity and the phenotype of ATM subpopulations under Rosi and ATM-Exosome treatment.
 
Overall design We performed a single-cell transcriptome analysis of stromal vascular cells of the epididymal fat isolated from obese mice treated for the last month with Rosiglitazone or with ATM-Exosomes or PBS Ctrl liposomes. Obese mice were fed with HFD for 10 weeks and were then treated for 4 weeks with Rosi-ATM-Exosomes (Rosi-ATM-Exos, n=3), HFD-ATM-Exos (n=4), or PBS Ctrl liposomes (n=3) (10^9 Exos twice a week) compared to Rosi diet (HFD containing 3 mg/kg/day Rosiglitazone, n=3).
