Summary |
Newly emerging transformed cells carrying genetic defects, including the active Ras mutant, are apicall extruded when surrounded by normal epithelial cells. This biological phenomenon is termed as cell competition. It remains elusive which factors prime RasV12-transformed cells to be expelled via cell competition. Here, We performe microarray analysis to search for molecules whose expression is changed in RasV12 cells surrounded by normal cells.

Overall design |
1.2×10^7 of 10:1 mix culture of MDCK and MDCK-pTR GFP-RasV12 cells or a single culture of MDCK-pTR GFP-RasV12 cells were cultured in collagen type I-coated dishes. After incubation with tetracycline for 24 h, GFP-positive cells were collected by an analytical flow cytometer, and total RNA was extracted from the isolated cells.
