Title Single cell sequencing data of islets from WT and Npc1KO mice

Summary Npc1 is a cholesterol transporter located on lysosomal membrane, and whether it is involved in regulating the function and differentiation of β cells is unclear. To understand molecular basis of the phenotype caused by Npc1 loss in β cells, we isolated islets from 2-week-old WT and Npc1KO mice for single-cell sequencing. We compared the gene expression profiles of different subpopulation of islet cells and found many changes of islet subpopulation and biological pathway occurred in Npc1KO islets.
 
Overall design To understand molecular basis of the phenotype caused by Npc1 loss in β cells, we isolated islets from 2-week-old WT and Npc1KO mice, digested the islet into single-cell suspension, and performed single-cell sequencing using 10X Genomics. Our islet samples were divided into two groups, WT and Npc1KO. Every islet suspension sample were digested by the islets of 3 same genotype 2-week-old female mice.
