Overall design |
The F. fujikuroi IMI58289 wild-type strain (FfWT) provided by Commonwealth Mycological Institute, Kew, UK, was used as a parental strain to generate the H2A.Z knock-down strain (Tetoff::FfH2A.Z), the constitutive H2A.Z overexpression (OE::FfH2A.Z) strain as well as the N- and C-terminal hemagglutinin (HA)-tagged H2A.Z (HA::FfH2A.Z, FfH2A.Z::HA) strains. Fungal mycelia were cultivated for three days in liquid ICI (6 mm glutamine) prior crosslinking with 1% formaldehyde. For immunoprecipitation, sonicated mycelia were incubated either with an anti-HA (Abcam, 9110), an anti-H3K27me3- (Active motif, 39155) or an anti-H3K4me3- (CellSignal, 9751) specific antibody. The assay was performed in biological duplicates, including input controls for every strain.
