Summary |
We wished to examine the effect that coculture with Desemzia incerta would have on the S. aureus transcriptional profile. We have previously shown D. incerta to have antimicrobial activity against S. aureus. We used a transwell coculture system to separate the two bacterial cultures. Transcriptional profiling revealed downregulated of genes in the arc operon, the pur operon, the mnh family proteins, and cap family proteins and upregulations in the arg operon and the ahp opeorn. Subsequent biological follow-up revealed decrease in S. aureus biofiom formation after D. incerta co-culture.

Overall design |
EGM139 D. incerta and USA300 Rosenbach MRSA were cocultured in a Corning 12 well transwell system as described in Wei et al. S. aureus RNA was extracted from the lower well using the ZymoResearch DirecZol RNA-Purification Kit. Sequencing was performed by SeqCenter. 4 replicates of S. aureus cocultured with D. incerta or TSB control were used
