Summary |
mRNA sequencing of mesenchymal stem cells transfected with YAP/TAZ siRNAs were treated with or without TNF-a for 24hr to profile gene expressions.

Overall design |
The siRNAs for human YAP, TAZ or negative control siRNA were chemically synthesized by FASMAC (FASMAC, Kanagawa, Japan). MSCs were seeded in 24-well plastic culture plates and maintained for 48 hr. Then, siRNAs (10 pmol/ml) were transfected into the cells by using Lipofectamine RNAi-MAX (Invitrogen, Carlsbad, CA). After 48 hr cultivation, MSCs were stimulated with or without 10 ng/ml TNF-a for 24 hr. RNA extraction was conducted by using RNA-iso (Takara). The construction and sequencing of the cDNA library were carried out by the Beijing Genomics Institute using the DNBSEQ platform. This project contains 12 samples of (3 from sicontrol-MSCs with non-treated, 3 from sicontrol-MSCs with TNF-a-treated, 3 from siYAP/TAZ-MSCs with non-treated, and 3 from siYAP/TAZ-MSCs with TNF-a-treated) RNA-seq data.
