Summary |
Due to the developmental abnormalities observed in SLMAP3 knockout mouse embryos, we aimed to investigated the impact of gene expression in E14.5 mouse embryonic fibroblasts (MEFs) isolated from these animals. The cells used for RNA-sequencing were previously immortalized with a transient expression of SV40 large T (SV40 1: pBSSVD2005 was a gift from David Ron - Addgene plasmid # 21826 ; http://n2t.net/addgene:21826 ; RRID:Addgene_21826). These immortalized cells express mostly the largest isoforms of SLMAP (~ 90kDa), which are the ones knocked out in the embryos, and therefore compensation because of other isoforms is negligible.

Overall design |
Immortalized mouse embryonic fibroblasts from 4 different wildtype and 4 different SLMAP3 knockout E14.5 embryos were used. RNA-sequencing was performed with each sample for the quantification of differential gene expression between the knockout and wild type cells.
