Summary |
Anti-CSF-1R treatment results in rapid regression of murne proneural glioblastoma models. However, the microenvironment response to treatment results in fibrosis that can promote tumor recurrence. We investigated the single-cell transcriptional profile of murine glioblastomas before and after treatment with an anti-CSF-1R inhibitor to determine the cause of fibrotic treatment response.

Overall design |
Gliobastomas were induced in mice using the RCAS model system at 5-weeks of age. Mice were monitored by MRI over the next 6 weeks until tumors developed that were at least 30 mm3 in volume. 3 mice were sacraficed prior to the initiation of treatment. 9 mice were treated daily with 200 mg/kg BLZ945 (Novartis), 3 were sacraficed 7-days post treatment, 3 at 28-days, 3 after rebound. All brain tumor samples were harvested and immediately embedded for cryosectioning for In Situ spatial transcriptomics using the 10X Genomics Xenium platform.
