Title Cut & Tag sequencing exhibited the chromosome binding regions of transcription factor IRF5 within murine bone marrow derived macrophages
Organism Mus musculus
Experiment type Other
Summary CUT&Tag technology represents a cutting-edge method for investigating protein-DNA interactions. Utilizing a hyperactive novel fusion enzyme (Hyperactive pG-Tn5 / pA-Tn5 Transposase), this technique, under antibody guidance, specifically targets and cleaves DNA sequences near target proteins. It boasts several advantages over traditional ChIP-Seq, including lower cell input requirements, enhanced signal-to-noise ratio, and improved repeatability. Employing this innovative approach, we examined the binding strength of interferon regulatory factor 5 in bone marrow-derived macrophages, and analyzed the biological behaviors of IRF5 following stimulation of these cells.

Overall design Cleavage Under Targets and Tagmentation (CUT&Tag) for the IRF5 binding regions (using anti-IRF5 antibody (abcam ab21689) in the murine bone marrow derived macrophages stimulated with CpG or control
