| Overall design |
His-MBP-tagged-HrAgo1 was heterologously expressed under the T7 promoter in E. coli strain BL21-Gold (DE3). His-MBP-tagged-HrAgo was purified using niNTA purification and size-exclusion chromatography. Nucleic acids were extracted from HrAgo1 and a small RNA library was generated and sequenced by GenomeScan (leiden, NL). Detailed methods are given in the associated publication.
