CLUSTER-NULL_ZT4
Organism |
Drosophila melanogaster |
Characteristics |
tissue: Fly heads genotype: w1118 ; mir959-962-KO
Treatment protocol |
Flies were collected at specified time point and frozen on dry ice, heads were removed
Growth protocol |
Adult flies were kept in constant 12 hr:12 hr light:dark conditions for 3 days before collection
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted from fly heads using TRIzol reagent (Invitrogen) as suggested by the supplier
Label |
biotin
Label protocol |
RNA was labeled for hybridization to Drosophila Genome 2.0 arrays (Affymetrix) using the IVT Express Kit (Affymetrix).
Hybridization protocol |
RNA was hybrized to Drosophila Genome 2.0 arrays following manufacturers recommendations. Arrays were washed and stained on a Fluidics 450 Station running protocol Midi_euk2 v3.
Scan protocol |
Drosophila Genome 2.0 arrays were scanned using a Affymetrix GeneChip Scanner 3000 and GCOS version 1.4.
Data processing |
Normalized log2 values were computed from the CEL files using the GC-RMA algorithm in the Bioconductor analysis package for R software (http://www.r-project.org)
