Treatment protocol Clams were notched in the shell next to the adductor muscles and injected with 100 µl of PBS. After stimulation, clams were returned to the tanks and maintained at 15ºC until hemocytes sampling.
Growth protocol R. philippinarum clams were obtained from a commercial shellfish farm (Vigo, Galicia, Spain). Clams were maintained in open circuit filtered sea water tanks at 15°C with aeration and were fed daily with Phaeodactylum tricornutum and Isochrysis galbana. Prior to the experiments, clams were acclimatized to aquaria conditions for one week.
Extracted molecule total RNA
Extraction protocol Total RNA isolation was conducted following the manufacturer's specifications in combination with the RNeasy mini kit (Qiagen) for RNA purification after DNase I treatment.

Label protocol Amplified and labeled according to the Agilent Low Input One-Color Microarray-Based Gene Expression Analysis protocol.
 
Hybridization protocol Hybridized according to the Agilent One-Color Microarray-Based Gene Expression Analysis protocol.
Scan protocol Array was scanned according to the standard Agilent protocol at a resolution of 5 microns modifying default setting to scan the slide twice at two different sensitivity settings (XDRHi 100% and XDRLo 10%).

Data processing Data were processed with Feature Extraction v9.5 and normalized by percentile shift at the 75th percentile using GeneSpring GX version 11.0.2 software (Agilent Technologies).
