Sample GSM1072637 Query DataSets for GSM1072637
Status Public on Jun 16, 2014
Title µ0.51 rep3 0 min
Sample type RNA

Source name chemostat 0.51 h-1 repet3 0min
Organism Lactococcus lactis
Characteristics strain: IL1402
treatment: µ0.51 rifampicin
time: 0 min
Treatment protocol At T=0 rifampicin was added into the culture at a final concentration of 500 µg.mL-1 so as to block transcription, mRNA levels were followed during the next 20 minutes
Growth protocol cultures were performed in chemically defined medium with low Branched-Chain Amino Acid concentrations. Chemostat cultures (growth rates of 0.51 and 0.11h-1) were isoleucine limited whereas isoleucine starvation occured progressively in batch cultures (grawth rates of 0.8, 0.38 and 0.04 h-1).
Extracted molecule total RNA
Extraction protocol see Redon et al. 2005. Role of mRNA stability during genome-wide adaptation of Lactococcus lactis to carbon starvation. J Biol Chem 280(43):36380-385
Label 32P
Label protocol see Redon et al. 2005. Role of mRNA stability during genome-wide adaptation of Lactococcus lactis to carbon starvation. J Biol Chem 280(43):36380-385

Hybridization protocol see Redon et al. 2005. Role of mRNA stability during genome-wide adaptation of Lactococcus lactis to carbon starvation. J Biol Chem 280(43):36380-385
Scan protocol see Redon et al. 2005. Role of mRNA stability during genome-wide adaptation of Lactococcus lactis to carbon starvation. J Biol Chem 280(43):36380-385
Data processing Raw data were processed to access mRNA concentrations and were normalized to the mean intensity of the reference membrane (before rifampicin addition) for each batch. mRNA half-lives (t1/2) were calculated from the degradation rate constant (k), which corresponds to the slope of mRNA concentration logarithm as a function of time with the relation t1/2=ln(2)/k
