Characteristics Monocyte derived dendritic cells were generated by culturing human monocytes isolated from buffy coats using CD14 microbeads (Miltenyi)
Treatment protocol cells were plated in fresh medium and incubated for 4h at 37°C before stimulation with 0.5 µg/ml S. abortus equi S-form LPS, pure TLR4 agonist (Alexis), together with 20 µg/ml CyP (Cyanobacterial Product) for 3h
Growth protocol CD14+ cells from PBMC were cultured for five days in 6-well plates in RPMI, 10% fetal bovine serum (Hyclone) supplemented with GM-CSF 50ng/ml and IL-4 1,000 U/ml. On day 5 monocyte-derived dendritic cells were harvested and replated at 0.5 106 cells/ml (1.5 106 cells/well).
Extracted molecule total RNA
Extraction protocol cells were harvested and lysed using TRIzol (Invitrogen).

Label protocol Affymetrix GeneChip Expression 3’ Amplification One-Cycle Target Labeling kit from about 5ug of total RNA
