Treatment protocol |
Whole blood was collected through venupuncture of the antecubital vein. White blood cells were isolated via red blood cell lysis and centrifugation. The cells were then stored in liquid nitrogen until RNA extraction. Extracted RNA was stored at -80°C.

miRNeasy kit (Qiagen) was used for extraction of total RNA and microRNA according to the manufacturer's instructions.

100ng of total RNA was prepared for hybridisation according to the standard Agilent protocol (microRNA Labeling & Hybridization Kit Manual, Agilent, Version 1.7, October 2009).

Hybridization protocol |
After fragmentation, samples were hybridized for 20 hr at 55°C to Agilent Human miRNA Microarrays (V3, release 16.0). Chips were washed and stained in the Agilent Microarray Hybridization Chamber SureHyb. The Agilent’s miRNA Complete Labeling and Hyb Kit was used according to Agilent's manual.

The chips were scanned on an Agilent G2565BA scanner.

The samples were between array normalized so that expression intensities were similar across all arrays using the quantile method, in the Partek Genomics Suite (v.6.6) application. Differentially expressed miRNAs were identified using a paired t-test.
