Treatment protocol Mammary glands were minced and digested with enzyme mix to generate single cell suspensions. YFP-marked luminal epithelial cells were sorted by FACS.
Growth protocol Mammary tissues were isolated from adult virgin female mice of their corresponding genotypes.
Extracted molecule total RNA
Extraction protocol RNeasy kit (Qiagen, #74134) was used according to manufacturer's protocol.
Label biotin
Label protocol Labeling, hybridization and scan performed by team of Dr. E. Fox at the Dana-Farber Cancer Institute Microarray Core Facility. The NuGEN Ovation V2 Amplification System was used to amplify and label the samples, according to manufacturer's protocol.
 
Hybridization protocol The biotinylated cDNA was added to a hybridization solution containing several biotinylated control oligonucleotides (for quality control), and hybridized to a microarray chip overnight at 45°C.
Scan protocol Each cDNA bound at its complementary oligonucleotide was excited using a confocal laser scanner, and the positions and intensities of the fluorescent emissions were captured.

Gene expression data for sorted wildtype luminal mammary epithelial cells marked by MMTV-Cre.
Data processing The raw CEL files were imported, normalized and analyzed with GenePattern analysis tools (dCHIP, GSEA) from the Broad Institute.
