After a 24h attachment period, the medium was discarded and replaced by incubation medium, consisting of Dulbecco's Modified Eagle's medium, supplemented with 50 mg/l gentamicin, 4 mg/l insulin, and 10-5 M hydrocortisone and containing 0 [vehicle control, 0.1% (v/v) dimethyl sulfoxide], 30, or 100 µM EMD.

Growth protocol |
Hepatocytes were plated in 60-mm dishes at a density of 3.5 X 10^6 cell/dish or in 6-well BD BioCoat plated (BD Biosciences, San Jose, CA) at a density of 1.5 X 10^6 cells/well in attachment medium and cultured under a CO2/air (5%/95%) humidified atmosphere at 37oC.

GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
