Primary CLL cells were cultured in RPMI supplemented with 10% fetal bovine serum (FBS), penicillin (50 U/ml), streptomycin (50 µg/ml) and recombinant human IL-4 (R and D Systems, Abingdon, UK) (5 ng/ml).

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA.

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45oC on Affymetrix U133 2.0 Gene Chips. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.

Data processing |
The data were analyzed with MADRAS bespoke software using Affymetrix default analysis settings and global scaling as normalization method.
