Treatment protocol Livers were removed after 64 days of supplementation from rats fed: 1) a standard diet (CT), n=7 arrays (7 female rats); 2) a rosemary extract (RE, 0.05% w/w) treated group, n=7 arrays (7 female rats).
Growth protocol Rat livers were rapidly removed after the animals sacrifice, rinsed and snap frozen in liquid nitrogen and stored at -80ºC prior to RNA isolation.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated from the rats livers using the RNeasy Plus Mini Kit (Qiagen, Madrid, Spain) (Ref.74134) following the manufacturer recommendations. RNA was eluted in RNase-free water, aliquoted and stored at –80ºC. Isolated RNA was quantified using the NanoDrop ND-1000 UV-Vis spectrophotometer (NanoDrop Technologies, Wilmington, DE) and the quality of the resulting RNA was determined by using the Experion RNA HighSens Bioanalyzer (Bio-Rad Laboratories, Inc.).
Label biotin
Label protocol RNA was processed according to the GeneChip® Whole Transcript (WT) Sense Target Labeling protocol (Affymetrix, Santa Clara, CA, USA). Amplified sense single-strand DNA was obtained using the Ambion® WT Expression Kit (Life Technologies). DNA was fragmented and labeled with the WT terminal Labeling kit.
 
Hybridization protocol Samples were hybridized for 16 h at 45ºC onto the GeneChip® Rat Gene 1.0 ST. Chips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000.

Gene expression data from liver isolated from lean female Zucker rat (fa/+). Standard diet + RE group. Time: 64 days.
Data processing The CEL files were used to extract and normalize the data using Robust Multichip Average (RMA) implemented with the algorithm RMA Sketch for 1.0 ST arrays in the GeneChip Expression Console software version 1.1.2 (Affymetrix).
