Treatment protocol HAOEC were seeded in a culture-insert on a flat clean surface until their attachment (Ibidi GmbH). Than, HAOEC were treated with 10 nM α-MSH or medium alone.
Growth protocol HAOEC were placed in a T 75 plastic flask at a density of 2500 cells/cm2 with EBM2 media plus supplements. All experiments were performed at cell passage 4-6.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the RNAdvance cell v2 kit (Agencourt) following recommended manufacturer's instructions. Then, total RNA was treated with DNase Turbo (Life Technologies) as described by the manufacturer's protocol. RNA quantification and purity were evaluated by micro-volume spectrophotometry using an Infinite 200 PRO plate reader (Tecan). The quality and integrity of the total RNA were checked by the RNA 6000 Nano Assay kit in a 2100 Bioanalyzer (Agilent Technologies).
Label Biotin
Label protocol RNA was used to generate biotinylated, linearly amplified RNA (cRNA) by Total Prep RNA Amplification Kit (Life Technologies), according to manufacturer's manual.
 
Hybridization protocol Standard Illumina hybridization protocol.
Scan protocol Standard Illumina scanning protocol for iSCAN.

Data processing Raw data were imported in the R software version 2.15.2 and analyzed with the lumi package from the Bioconductor Suite version 2.11. Variance stabilizing transformation (VST) was applied and transformed data were then normalized by robust spline normalization (RSN).
