Fish (n=4) were intramuscularly injected with 1,5 µg of the plasmid constructs containing the gene sequences of SVCV or VHSV glycoproteins or the empty plasmid. After three days muscle samples were excised from the site of injection and processed from RNA isolation and gene expression analysis with microarrays.

RNA was isolated using the E.Z.N.A HP Tissue RNA kit (Omega bio-tek) according to intructions provided in the kit. RNA was kept at -80 oC until used.

Each amplified and labelled sample was hybridized at 65 oC for 17 hours.

Data was analysed using GENESPRING software (GX v. 11.0.).Signal intensities for unique probes within a probe set were averaged to obtain an expression value (median) for the probe set (Gene-level analysis). expressed was defined as transcripts with a log2-transformed expression value greater than 1.
