Adult female mosquitoes were collected at 3 h after regular blood feeding (samples #3-4) and P. berghei-infected blood feeding (samples #7-8), as control females fed on 10% sugar solution were used (samples #1-2). Samples #5-6 were prepared using adult female mosquitoes injected with 0.6mg of dsRNA with targeted activity against Ago1 and Ago2.

Total RNA was isolated from 30-50 females using a TRI Reagent (MRC).
small RNA libraries were prepared using Illumina’s protocol (Truseq small RNA, # 15004197 Rev. C). 2mg of total RNA was used to construct small RNA libraries.

Basecalls performed using CASAVA version 1.8.2
Data were processed using miRDeep2 package (2.0.0.5). The raw reads were firstly trimmed against Illumina adapters, then mapped to the AgamP3 genome assembly with the Mapper module.
The miRBase release 19, An. gambiae annotation was refined to contain both 5p and 3p mature sequences and then used for the quantification.
