KLSCD34- stem cells were sorted on a FACS Aria (BD Biosciences) into buffer RLT (Qiagen) containing 2-ME and total RNA was extracted by trizol.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 100ng total RNA.

Hybridization protocol |
Following fragmentation, 2ug of cRNA were hybridized for 16 hr at 45C on mouse 1.0 ST Gene Array. Hybridization was performed using an Affymetrix FS450 hybridization oven. GeneChips were washed and stained in the Affymetrix Fluidics Station FS450.

Scan protocol |
GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G with AGCC.

Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
