Extraction protocol Total RNA (plus miRNAs) was isolated using a modification of the RiboPure™-Blood kit from Ambion (Austin,TX) according to the manufacturer’s protocol. The concentration of total RNA and integrity were determined by using Nano-Drop ND-1000 Spectrophotometry (NanoDrop Tech, Rockland, Del) and gel electrophoresis respectively.

Hybridization protocol hybridized on miRCURY LNA™ Arrays, using MAUI® hybridization system
Scan protocol Chips were scanned on Innopsys Scan 700, microarray scanner at 532 nm
Description non-heart failure subject
Data processing miRNA microarray data were analyzed by subtracting the mean background signal (B532nm) from the test signal (F532nm). All the 3 versions (v12; v16; 6thGEn chips and v18; 7thGen Chips) of Exiqon miRNA microarray probes were matched to the v18 miRNA name. Subsequently the collated data was analysed for further interpretations.
Raw signal values in the non-normalised matrix and quantile normalised data in the normalised matrix spread sheet respectively. The quantile normalization of the probes were carried out on PartekÒ Genomics Suite™ 6.6 software.
