Title |
Pax3-injected animal cap, stage 12, induced with Dex, replicate 1 |
Sample type |
RNA |
Source name |
Pax3-injected animal cap_stage 12_induced with Dex
Organism |
Xenopus laevis |
Characteristics |
tissue: Xenopus laevis animal cap explant, stage 12 dex treatment: yes pax3 injection: yes zic1 injection: no chx treatment: no biological eplicate: 1
Treatment protocol |
Cycloheximide (0,1mg/ml) was then applied to the healed animal caps, from stage 10 to 10.5 (i.e. for 30 min at 23*C), then dexamethasone was added at stage 10.5 to the cycloheximide-containing medium. Explants were rinced and lysed after two additional hours at 23*C, i.e. when sibling embryos reached stage 11.5-12.
Growth protocol |
Xenopus laevis 2-4 cells stage embryos were injected with inducible Pax3-GR+/-Zic1-GR constructs and then grown at 15°C in 1/3 MMR until stage 9
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated using the standard trizol protocol and eppendorf phase-lock gel tubes, followed by proteinase K digestion, and Qiagen Rneasy columns
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 ug total RNA
Hybridization protocol |
Hybridization was completed using the standard affymetrix protocol
Scan protocol |
Scanning was completed using an Affymetrix Genechip scanner 3000 7G
Description |
Pzc1; targets_1G.CEL
Data processing |
The data was analysed with R (v.2.14.2) and the Bioconductor Biobase (v.2.14.0) affy library (v.1.32.1 package) with default settings.
