Following excision, the tissue samples were immediately immersed in RNAlater, (QIAGEN) and stored at −20°C until RNA extraction. miRNAs were purified using the miRNeasy mini kit (QIAGEN).

Total RNA 100 ng were subjected to cDNA synthesis by Ambion WT Expression Kit (Life technologies). Obtained 5.5 μg cDNA were biotinylated and then fragmented by GeneChip WT Terminal Labeling Kit (Affymetrix).

Hybridization protocol |
After creating the hybridization cocktail, biotinylated cDNA were hybridized with GeneChip Human Exon 1.0 ST Array (Affymetrix) for 17 hours.

Scan protocol |
The arrays were scanned using the Affymetrix Gene Chip Scanner 3000.
