U251 cell line
Organism |
Homo sapiens |
Characteristics |
sirna transfection: siRNA targeting TLX stimulation: IFN-gamma cell type: astroglioma cell line: U251
Treatment protocol |
After cells had settled and adhered in 6-well plates, they were transfected with 10 nM siRNA targeting human TLX (or negative control siRNA targeting luciferase) using HiPerfect transfection reagent. 72 hours after transfection, cells were stimulated with 10ng/ml of recombinant IFN-gamma for 8 hours
Growth protocol |
U251 cells were grown to subconfluency in 6-well plates with DMEM media supplemented with 10% serum and antibiotics.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted from individual wells of 6-well plate using the Qiagen Rneasy Plus kit.
Label |
biotin
Label protocol |
Biotin labeled complementary RNA (cRNA) was made using the EpiCentre Nano Total-Amp kit
Hybridization protocol |
Standard Illumina hybridization protocol
Scan protocol |
Standard Illumina scanning protocol
Data processing |
The data were quantile normalized without background subtraction using Illumina GenomeStudio V1.8
