Figla null mouse line was made by targetted disruption of Figla gene in the mouse chromosome by homologous recombination in mouse 129Sv ES cells. The correctly targetted ES cells were injected in the blastocyst of C57BL/6 mice

total RNA was extracted from the normal and homozygous null mice by using RNeasy mini kit (Qiagen, Valencia, CA). homozygous null mouse RNA was confired by one step RT PCR using primers for ZP2 and MSY gene and absence of ZP2 and presence of MSY gene was considered as homozygous null ovary RNA. Poly (A)+ RNA was isolated with oligo d(T)- conjugated magnetic beads (Dynal- Invitrogen, Carlsbad, CA). 5 microgram of this RNA was used for construction of these libraries using the original SAGE protocol of Velculescu et al (Velculescu et al, 1995). Final concatenated ditags were cloned into Bluescript pKS (Stratagene, LA Jolla, CA) using BamHI site for blue white screening. Sequencing was perfomred at NIH Intramural Sequencing Centre (NISC). Low quality sequence data was eliminated by Phred analysis
