Title |
CK-24 h-rep3 |
Sample type |
RNA |
Source name |
The internodes meristem after normal-water sprayed treatment 1 day (24 h)
Organism |
Zea mays |
Characteristics |
tissue: internodes meristem
Treatment protocol |
At the jointing stage (8 leaves), 600 mg/L ethephon was sprayed on 100 seedlings served as the treatment group, and a normal-water sprayed on another 100 seedlings served as the control group, respectively.
Growth protocol |
Sterilized seeds of maize (Zea mays) were sown in a greenhouse (temperature, 28°C day/20°C night; 16 h photoperiod; 40% relative humidity) at China Agricultural University, Beijing.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 200ng total RNA (3' IVT Express Kit User Manual, 2008, Affymetrix).
Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45°C on GeneChip Wheat Genome Array in the Affymetrix Hybridization Oven 640. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
GeneChips were scanned using the affymetrix scanner 3000 7G.The scanned images were first assessed by visual inspection then analyzed to generate raw data files saved as CEL files using the default setting of AGCC.
Description |
Gene expression data.
Data processing |
GeneChip Operating Software (GCOS1.4, Affymetrix), dChip 2008 (Li C, 2001aLi C, 2001b), and MAS 5.0.
