Treatment protocol All the specimens were collected from intervertebral discs of cadaveric donors or patients within 3 hours after anatomy or surgery respectively, following rinsed with phosphate-buffered saline, the nucleus pulposus tissue was carefully collected under stereoscopic microscope.
Extracted molecule total RNA
Extraction protocol Control and degenerated samples were prefrozen with liquid nitrogen, ground into powder with mortar, extracted with Trizol reagent according its protocol.
Label Cy3
Label protocol 1ug of total RNA was reverse transcripted into cDNA with T7 Promoter Primer and MMLV-Rtase, and then was labled with T7 polymerase and Cyanine-3-CTP.
 
Hybridization protocol The labeled cRNAs were purified with RNeasy mini column and subsequently hybridized onto the Human LncRNA Array v2.0 (8 x 60K, Arraystar).
Scan protocol After having washed the slides, the arrays were scanned by the Agilent Scanner G2505C (Agilent Technologies).
Description Derived from the lumbar disc of cadaveric donor with no significat degenerative signs on magnetic resonance imaging
Data processing Agilent Feature Extraction software (version 11.0.1.1) was used to analyze acquired array images. Raw signal intensities were normalized in quantile method by GeneSpring GX v11.5.1 (Agilent Technologies), and low intensity LncRNAs and mRNA (at most 4 out of 10 samples have flags in Present or Marginal ) were filtered.
GeneSpring GX v11.5.1 (Agilent Technologies) computed normalized signal intensity. Log2 value of normalized Intensity of each sample were presented in matrix table.
