Treatment protocol Single cells were dissociated from MCF7 spheres and treated with 10nM phtpp or dpn in triplicates for 48h. Monolayer MCF7 adherent cells were treated with 10nM dpn in triplicates for 48h.
Growth protocol MCF7 spheres were cultured in the selective medium for 3 generations. MCF7 adherent cultures were maitained in white DMEM supplied with 5%FBS
Extracted molecule total RNA
Extraction protocol Total RNA was extracted by using RNeasy mini kit from QIAGEN according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated sense-strend DNA is generated from 100ng of total RNA using Affymetrix standard protcol(WT PLUS reagent user manual)
 
Hybridization protocol 5ug of target is hybridizedto HTA 2.0 Array in Affymetrix Hybridization Oven 645 for 16 hr at 45C
Scan protocol Arrays were scanned using the Affymetrix Gene Chip Scanner 3000 7G
Description Gene expression data from MCF7 spheres treated with DPN.
RM01_D1_MCF7_S
Data processing Gene level normalization was done using Robust MultiChip Analysis (RMA) algorithm in Affymetrix Expression Console. Differentially expressed genes between treatment groups were identified by one-way Analysis of Variance (ANOVA) of preprocessed data in Affymetrix Trancriptome Analysis Console, including genes with±1.2 fold change, P-value of <0.05 and known gene annotation.
