Treatment protocol |
Mice were treated with either saline control or 5-Aza-2'deoxycytidine (0.2mg/kg/day) by daily I.P. injection for 3 weeks. After 2 weeks of treament, 3 out of the 4 caudal branches of the left carotid artery (LCA) of mice were ligated while the right carotid artery (RCA) was left untouched as the contralateral internal control. After 1 week of ligation, mice were sacrificed by CO2 inhalation.

C57Bl6 mice were fed normal chow diet for 9 weeks.

Extraction protocol |
Mice were perfused with saline and carotid arteries were extracted and flushed with 150 ul of Qiazol. The Qiagen miReasy Kit was used to extract and purify total RNA. Quality control was performed with Agilent Bioanalyzer.
