Treatment protocol The media was switched to Ham’s F10 supplemented with 10% horse serum and transfected with 10 µM AMO as indicated for 48 hours. The media was then replaced with Opti-Mem and cells re-transfected with 10 µM AMO with 50uM PDGF-AA or control. After 48 hours, total RNA was extracted.
Growth protocol Cells were grown for 2.5 days in DMEM supplemented with 10% FBS prior to treatment.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the RNeasy Mini Kit as per the manufacturer’s instructions (Qiagen).
Label biotin
Label protocol Biotinylated single stranded cDNA were prepared according to the standard Affymetrix protocol from 100 ng total RNA.
 
Hybridization protocol Fragmented biotin-labeled single stranded DNA was hybridized to Affymetrix Mouse Gene 1.0 Array according to the standard Affymetrix protocol. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol GeneChips were scanned using the Affymetrix GeneChip Scanner 3000 7G.
Description Gene expression data from WT fibro/adipogenic progenitors isolated from a noninjured animal and treated in vitro with Intronic PDGFRα 5' splice site AMO and 50uM PDGF-AA.
Data processing The data were analyzed with the Affymertix Expression Console software using Affymetrix default RMA analysis settings.
