Treatment protocol Purified human B cells were isolated by negative selection from PBMC using the human B Cell Isolation Kit II, human (MACS, Miltenyi Biotec, San Diego, CA) according to manufacturer’s recommendations. SLCL and LCL lines were treated as follows: cells were washed, resuspended at a concentration of 5x106/mL in complete media consisting of RPMI 1640 (Fisher Scientific of Life Technologies, Foster City, CA) supplemented with 10% fetal calf serum and 1% penicillin/streptomycin and incubated at 37*C. After one hour, complete media was added to achieve a final concentration of 0.5x106 cells/mL. Cells were then placed in a 37oC CO2 incubator for 12 hours before collection by centrifugation and lysis in TRIzol Reagent (Life Technologies, Foster City, CA) for isolation of total RNA.
Growth protocol SLCLs and LCLs were maintained in complete media consisting of RPMI 1640 (Fisher Scientific of Life Technologies, Foster City, CA) supplemented with 10% fetal calf serum and 1% penicillin/streptomycin and incubated at 37*C.

Data processing The resulting Ct values (Matrix non-normalized) were normalized first to the median Ct value of the individual array (Matrix normalized) and then to the DCt of the miRNA from the control samples, to produce the DDCt (Fold Change). All are divided by the control (BCELL).
