Title |
Microglia_specificpathogenfree_6hr_after_LPS_ic_3 |
Sample type |
RNA |
Source name |
Microglia, LPS i.c., SPF
Organism |
Mus musculus |
Characteristics |
cell type: microglia treatment: LPS i.c. condition: specifc pathogen free (SPF) age: 6-8 weeks old strain: C57BL/6
Treatment protocol |
5 µg LPS were diluted in 10 µl PBS and applied intracranially under isofluoran anesthesia. Animals returned to their cages once they had recovered their righting reflex. 6 hours later, animals were analyzed. Control animals were injected with 10 µl sterile PBS i.c.
Growth protocol |
Mice were housed under SPF or germ-free conditions. Animals were 6-8 weeks old.
Extracted molecule |
total RNA |
Extraction protocol |
Adult microglia were harvested using density gradient separation. CD11b+CD45lo microglial cell populations were FACS-sorted and collected directly in cell lysis buffer and subsequently RNA was isolated with the Arcturus Pico Pure RNA Isolation Kit (Life Technologies) according to the manufacturer's protocol.
Label |
Biotin
Label protocol |
Total RNA was amplified using the Ovation PicoSL WTA System V2 and further processed using the Encore Biotin Module according to the instructions supplied by the manufacturer (NuGen).
Hybridization protocol |
Biotinylated cRNA was hybridized on GeneChip® Mouse Gene 2.1 ST Arrays , that were stained, washed and scanned following standard procedures.

Description |
Microglia from specifc pathogen free (SPF) mice treated with LPS i.c.
Data processing |
Background correction and normalization was performed using the RMA-Sketch implementation in Affymetrix Expression Console
