Title |
A673 treated with Puromycin for 5 days with concentration of 0.09 uM (EC50), biological rep3 |
Sample type |
RNA |
Source name |
A673 cell line, a human EWS/FLI translocated Ewing sarcoma cell line.
Organism |
Homo sapiens |
Characteristics |
A673 cell line that has been treated with Puromycin for 5 days with concentration of 0.09 uM (EC50).
Growth protocol |
The Ewing sarcoma cell line A673 was grown in Dulbecco's Modified Eagle Media (DMEM) (Cellgro) with 10% Fetal calf serum (FCS) (Sigma Aldrich) and 2 mM sodium pyruvate (Cellgro) at 37C with 5% CO2.
Extracted molecule |
total RNA |
Extraction protocol |
RNA was extracted with Trizol per the manufacturer's protocol (Invitrogen) and prepared for hybridization to Affymetrix high-throughput arrays.
Label |
biotin
Label protocol |
Two micrograms of RNA was used for target preparation as per the Affymetrix Gene Chip Automated Station (GCAS) protocol (Early Access User Manual Version 1.0).
Hybridization protocol |
3.5 ug of cRNA was hybridized to Affymetrix 96-well Gene Chip HT human array with standard hybridization, blocking, washing, and staining per the Affymetrix Gene Chip Automated Station (GCAS) protocol (Early Access User Manual Version 1.0).
Scan protocol |
Scanned per the Affymetrix Gene Chip Automated Station (GCAS) protocol (Early Access User Manual Version 1.0).
Description |
Gene expression data from A673 cell line that has been treated with Puromycin for 5 days with concentration of 0.09 uM (EC50).
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix's default analysis settings. The samples in the data set have been scaled so that the mean expression of all genes for each sample is equal to the mean expression of all genes for the median sample.
