Title |
reproductively inactive turkey rep 3 |
Sample type |
RNA |
Source name |
Pituitary gland of reproductively inactive turkey
Organism |
Meleagris gallopavo |
Characteristics |
gender: Female developmental stage: Adult reproductive stage: Photorefractory
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using RNeasy Lipid Tissue Mini Kit (Qiagen) following manufacturer’s instructions
Label |
Alexa fluor 555, Alexa fluor 647
Label protocol |
cDNA was synthesized and fluorescently labelled using SuperScript Plus Indirect cDNA Labeling System (Invitrogen) following manufacturer’s instructions
Hybridization protocol |
Hybridisation was performed at 42°C for 16 hr using SlideHyb Buffer (Applied Biosystems) in hybridization chambers. The arrays were washed with increasing stringency using salt sodium citrate and immediately scanned
Scan protocol |
The arrays were scanned on a ScanArray 5000 (Packard BioScience)
Data processing |
The fluorescence intensity of each spot from the images was extracted using BlueFuse software (BlueGnome Ltd.) and exported to JMP Genomics (SAS Institute Inc.). The raw intensity was first transformed to log-base 2 and performed within-array Loess normalization and then standardized to a mean of 0 and standard deviation of 1 across arrays.
